In the Heights volgt verschillende personen woonachtig in de New Yorkse buurt Washington Heights. Usnavi is eigenaar van een bodega en helpt zo nu en dan zijn Cubaanse buurvrouw op leeftijd. Hij heeft een oogje op de vrouw die in een schoonheidssalon werkt en droomt van een leven in de Dominicaanse Republiek, waar tevens zijn roots liggen. Intussen keert zijn jeugdvriendin Nina terug naar de buurt. Zij heeft haar ouders iets belangrijks te vertellen.
